My advice is to start off with a character steeped in the lore you know/like most. That said, I think it would also be a good idea to start with a character that is Neutral with respect to Good and Evil.
Sigil's a setting with LOTS of gray, and starting off with a Good character can be challenging when you have demons, devils, aberrations and undead wandering freely through the streets and mixing with mortals and upper planar beings.

Well, what I would do is figure out what Faction sounds cool to you. I originally started with a Bleak Cabal warlock. I didn't like that, so I switched it up to an Athar warlock. (New character.) First thing is, check out the Wiki. Look at races, Factions, and classes. Keep in mind, because people are busy the wiki isn't 100% up to date.
While looking at races and classes, note how the different races have different ECLs ( http://sigil-nwn2.wikia.com/wiki/Races_ecl ) and some of the template classes add ECL ( http://sigil-nwn2.wikia.com/wiki/Specia ... ext=Search ). That's important, because high ECL races don't usually make good spellcasters.
Then, I'd check out some external Faction lore resources. Here's a few:
Planeswalker Handbook -- 2E PHB for the setting
Player's Guide to Planescape -- 5E guide for the setting; keep in mind that not everything applies due to NWN2 being 3.5E. It's basically a conversion from 3.5E, so most of it still works.
Factols Manifesto -- Lore relating to higher level Faction abilities
In the Cage: A Guide to Sigil -- Sigil Overview
Planescape Campaign Setting -- Self-explanitory.
Background is also important. Did you come from a Prime world (like Toril, where the Sword Coast is; or maybe Oerth or Golarion)? Or did you come from an Inner Plane (the Abyss, Nine Hells, Mercuria, etc.)? Planars and Primes tend to act different and have different motivations.

G'day everyone,
I've been poking around your forums and wiki for a couple of days now and I'm rather intrigued to give your server a try for once. I'm a BGTSCC vet (don't hold that against me please!) so I'll be bringing 5+ years of NWN2/DnD RP experience with me.
My only problem so far has been the character creation page! So many choices! So many possibilities!
Where I would appreciate some help is if certain factions/groups are in need for certain character/class type to bolster their ranks, preferably late night Western USA time players (I am GMT+10 but I get off work most days around 9-10pm WST, give or take).
I tend to play Good characters but I wouldn't mind trying an evil character as my main here. I am currently tossing up a few options: shadow dragon, gauth beholder or illithid, but I also have no qualms playing a slave class eg. mentally dominated troll frezerker, hill giant, even a kobold or goblin. I also tend to gravitate towards casting classes (I'm a rather experienced cleric RPer) so keep that in consideration with your suggestions.

First off, welcome to the City of Doors :)
I wouldn't worry too much about niche's. Take your time with picking a Faction, find something that fits what -you- want to play. That's more important than filling gaps, especially since RPing in a Faction is so directly tied to your characters desires and goals.
If you need a run down of the Factions though, just give a shout and I'll be happy to give you an overview :) (as I'm sure others might as well!) .

Like Daggerknight said, don't join a faction to fill a gap. Factions are so integral to philosophies and beliefs, and even without a ton of PCs in them they're still major political movers and shakers. You're more than welcome to simply not pick one, too.
Player Groups aren't as limiting, but they also have zero political power. Don't expect to make any big changes by joining a Player Group. No matter how big the group becomes they're never going to have their own representation in the Hall of Speakers. Aside from having access to group resources and having some people who will stand with you when push comes to shove there isn't really much of a reason to be in one. But if you really like the group and what it has going on there's also no inherent reason not to join them.
Oh! And welcome to the cage! Watch out, there's puddles of blek everywhere.

The general guideline is "start simple, get to know places" and that'll help you figure out what you'll want to play in longterm. A clueless (term for people coming from prime material planes such as Toril) fighter is an ever popular starting kit, you can ask stupid questions without feeling awkward and punch people who are annoying. Practically all you need in life.
It's as much about getting used to the server as it is with getting used to the players and groups. After you know the places and people, it's much easier to make a more "targeted character".

I'm usually the kind of person that likes to get saturated in lore first (already read through Sigil Cant and Planescape Cosmology) and then create a character accordingly. My problem is that there is such a great variety of options for characters that I seriously cannot choose what I want to play!
I could, of course, go with what I know best (Forgotten Realms humanoid) but I'm hoping to broaden my perspectives a bit with something a little more interesting.
I would like to read a good overview on the factions but the link provided in the Existing Factions & Possible Guilds pin seems to be broken.
I also noticed Gem dragons (and dracolich) on the wiki but they are not available at character creation: I assume these are not yet implemented?

Yes, the gem dragons are still being developed and will be implemented soon.
(As an aside, although you are perfectly free to create any character you want I would not advise making a character from Toril if you want to fill a niche. That niche is very full already. ^^;;)
